Midi does not contain any sounds, it's pretty much sheet music compressed into one file. I used a website and uploaded the mp3 and it converted it to work in synthesia, but it only did it on small files. Ableton Live will convert audio to MIDI. It's shaky with really complicated program information but it gets the job done in most cases!
If I didn't get this wrong, OP wants to extract audio from MKV, then the video stream, and use them separately in a video editor. Try FFmpeg if you are comfortable with a command line tool. To extract the audio stream: ffmpeg -i input.mkv -vn output.mp3. To save the video stream: ffmpeg -input.mkv -an output.mp4.
Im thinking of tidying up a bit and may convert the chapterised mp3s into single files to reduce the number of files I have to deal with. Previously ive tended to splice them into a single mp3 file using makeitone. The problem is this loses the chapter info. As more players now seem to support m4b im now thinking of converting the mp3 to m4b.
